Actor Vince Chase is about to have his latest film Head On open across the country. With him along for the ride are his crew, his brother Johnny Drama and his best friends, Eric and Turtle, who've known him since his early days in Queens. In the opener, Eric ends up clashing with Vince's superagent, Ari, who thinks he has found the perfect project for him. After Eric convinces Vince that the film is a bad move, Ari goes ballistic.

Vince has himself a new girl named Fiona and the rest of the crew is not to happy about her because she's trying to change Vince's routine around and doesn't want him to hang so much with the guys. While at the gym, Eric is given a script by another agent, and urges Vince to take the project because it's set in Queens. Meanwhile, Drama is concerned about all of his friends' relationships.

Vince wants to make Queens Boulevard, but Ari has yet to close the deal and is pitching him other projects that Vince wants no part of. It turns out that the agent who gave Vince the script is Ari's rival, Josh Weinstein, and he decides to invite the guys to a beach party. Meanwhile, Turtle makes fun of Gary Busey's paintings while visiting an art gallery and Drama finally gets representation, but is unhappy with the outcome.
As the pre-production of Queens Boulevard is underway, Vince is fond of the indie director hired, but Eric has some major concerns, especially when he finds out that a scene has been added which will put Vince in an awkward position. Meanwhile, Drama gets some extra money courtesy of a residual check and Turtle wants some of it to spend on "dancers". Also, Ari decides it's time to mix business with some pleasure.
Eric decides it's time for him to have an official title, as he is tired of being known as "Vince's Guy". As Vince prepares to say good-bye to L.A. as the shoot for Queens Boulevard will take three months in New York, Turtle decides to throw him a party. Meanwhile, Drama finally gets a major audition for a TV show.
Now that his latest film, Queens Boulevard is completed, Vince, Eric, Turtle and Drama all return to L.A. to pick up where they left off. Eric is trying his hardest to be the best manager he can for Vince and trying to deal with his relationship with Kristen. Turtle feels unappreciated now that Eric has this new role. Meanwhile, Ari has a new project for Vince, who is less than thrilled with what it is, director James Cameron's new film Aquaman.
Ari continues trying to lure Vince into doing the Aquaman project, but Vince would rather spend the time at Jaime Pressly's beach house. While there, Eric ends up with a model and feel extreme guilt for cheating on Kristen, who cancelled their last date. Meanwhile, Drama is concerned over his body (especially his legs) and wonders if he will ever get better roles.

After the guys get kicked out of their mansion, Vince falls in love with one of Marlon Brando's old homes but when he realizes it's out of his league, he begins reconsidering the Aquaman project. Soon the crew is invited to the Playboy Mansion, all but Drama, who has been banned for life by Hugh Hefner himself. However, Drama quickly finds a way to get let in, with "help" from Pauly Shore.

Eric worries as to why the Aquaman deal is not yet in writing. However, Ari is trying is best but just cannot close the deal, but soon discovers that Vince is no longer the front runner for the project. Vince, with help from Turtle, decides it's time to decorate the new mansion as he soon receives good news about Queens Boulevard. Meanwhile, Drama is still concerned about his calves and is considering a $10,000 surgery to augment them.

This causes both James Cameron and Eric to worry about how the production of Aquaman will turn out. Meanwhile, the crew has to shop for Ari's daughter's Bat Mitzvah and end up meeting the daughter of Ari's boss. Soon both Eric and Vince show interest in her.

Ari gives Vince a painting, but quickly realizes it's a fake and goes out of his way to give him the original one. Drama gets too over-excited about working with his latest co-star, Brooke Shields. Eric's relationship with Sloan begins to affect his professional life.

Vince's relationship with Mandy intensifies and Eric is forced to deal with both of their agents, who are not happy about the situation. Ari tries to lie to Eric about something James Cameron said about Vince's behavior. Meanwhile, Turtle decides to promote a new track for the Queens Boulevard soundtrack after the crew discovers a new artist.
After seeing Mandy with her ex, the crew begins to worry about Vince because of how much he has professed his love for her. Drama and Turtle decide to tail Mandy and get photographic proof of her rendezvous. After Mandy finds out, she tells Vince how she really feels about him. Meanwhile, Ari finally hits a breaking point with Terrance and ends up in a hairy situation.
After Vince's mood deteriorates, he begins alienating himself from the crew and considers dropping out of Aquaman. This causes them to begin pondering their futures without him with them. Meanwhile, Turtle finally gets Saigon a showcase as Drama ends up being dropped by his agent. Eric ends up getting offered an agent job by Terrance, now that he has fired Ari, who is trying to get back into the game.

Drama is informed that he's going to be a target on a new Pauly Shore prank show. Eric tries to ease the tension between Vince and Amanda. Meanwhile, Ari becomes envious toward a former frat brother.
Medellin is once again on the table for Vince, but Ari and Nick Rubenstein have until sundown on Yom Kippur to seal the deal. Meanwhile, Drama forms a unique bond with a race horse.
Eric and Vince try to get Medellin up and going, but the rights to the screenplay trip them up. Drama, stressing over the Five Towns premiere, finds relief at a massage parlor. Meanwhile, Turtle meets the perfect girl for him -- they both share a mutual love for limited-edition sneakers -- but her father gets in the way of his game.
Drama gets an offer for a role in Rush Hour 3. Ari is able to finally find a financial backer for Medellin, but the backer's intention towards Vince are shady at best. Meanwhile, Turtle has a date with Kelly with her father as their chaperone.

Eric gets into a minor car accident with Anna Faris and thinks that they shared "a little moment" in the process. Mrs. Ari, much to Ari's chagrin, wants to reprise her role on The Young and The Restless.
Ari is forced to work overtime when Billy delivers his script for Vince's next film; Eric has his hands full with a new client and her boyfriend.
The guys' trip to Cannes for the “Medellin” premiere is hampered by a security threat at the airport; Ari deals with his wife's and Lloyd's desires to go to France; Billy does his best to accommodate Eric's new client.
In the fourth-season finale, attempts to sell “Medellin” at Cannes are complicated by the fact that no one has seen the finished product. Meanwhile, the popularity of “Viking Quest” gives Drama a boost.
As dismal reviews for Medellin continue to roll in, Vince is hiding out in a Mexican paradise with Turtle. Meanwhile, producer Carl Ertz wants a meeting with Vince about a role in a guaranteed hit, and Eric and Ari go all out to persuade him to come back to LA. Drama turns prima donna during a photo shoot for Five Towns,and kindles his "virtual" romance with his Cannes girlfriend, Jacqueline.

The guys step up their security after a suspicious break-in. After Ari steals teen heartthrob Zac Efron from Adam Davies, Davies counters by offering Lloyd a job as an agent. Meanwhile, Drama follows up on an insider tip on Vince's suspected stalker and Ashley grows increasingly suspicious of Eric.
Drama eyes a part on Melrose 2009 and asks Lloyd if he can get him out of his current gig on Five Towns. Still irate over Lloyd's job offer, Ari goes to war against Lloyd to get Drama out of his contract. Citing their trust issues, Ashley asks Eric if she can read his emails so that they can have an open and honest relationship. Turtle feels guilty about his love triangle, of sorts, with Jamie-Lynn and a UCLA co-ed.
Eric visits his doctor's office after getting some unsavory news from Drama about his latest fling. Ari investigates Terrance after he unexpectedly offers to sell Ari his share of his agency. Meanwhile, Turtle prepares for Jamie-Lynn's departure and Drama's emotions get the best of him during his audition for Melrose 2009.
Season Six Finale. Eric plans a special lunch for Sloan that doesn't go as smoothly as he hoped. Ari tries to convince his wife that buying Terrance's company is not a personal vendetta. Meanwhile, after repeated attempts to get over Jamie-Lynn, Turtle decides to make a grand gesture to be with her; Drama, resolving to give up acting for good, turns down a chance to re-audition for Melrose 2009; and Matt Damon, with help from Lebron James, strong arms Vince to get involved in his charity before Vince leaves for Italy.
