The episode begins with two men working in the sewer, who are frightened by the appearance of a humanoid lizard. They climb up the ladder to get away from him, and while one gets away successfully, the other is caught by the foot through a storm drain and pulled under. The other sewer worker drives off, frantically, while having hallucinating images of the Lizard. Spider-Man catches him before he kills someone, then tells him not to “blame this one on me”, and then web-slings off.

We now see Peter at the Daily Bugle, in which J. Jonah Jameson explains that there’s been sighting of a giant lizard, which Peter believes is just a hoax. The Lizard is believed to be created by neogenics, in which Peter is an expert on. Eddie Brock, who was assigned the story, is dropped for Peter, as Jameson wants Peter on it due to him understanding neogenics. Peter still admits that he believes that it’s a hoax, until he’s offered one thousand dollars to get a picture of the Lizard.

Peter takes the bus home, pondering what he’ll buy with the thousand dollars, should he obtain a picture of the Lizard. Once he enters his home, in which he’s staying with his aunt May, he notices that she’s been hiding bills from him that she can’t afford to pay. Peter decides that he’ll use the money to aid Aunt May and get her caught up on her bills.

He changes into Spider-Man, then heads to the sewer where the first sighting occurred. He notices giant, reptilian foot prints, which he photographs. He decides to take the photographs to Dr. Connors, when he runs into Debra Whitman, who tells him that whatever the creature was, it most likely walked upright. Once they get into ESU, we see Eddie Brock, who’s decided to go against Jameson’s wishes and continue working on the story to attain a photograph of the Lizard, as well. Peter’s Spider-Sense goes off, which motivates him to run to Dr. Connors’ lab, in which he sees the Lizard with the neogenic recombinator. Debra has followed him, and disturbs the Lizard, who hits her in the chest, thus knocking the wind out of her. The Lizard jumps out the window, and Peter tries to follow him, though when he’s about to change into Spider-Man, Debra regains consciousness, so he knows he can’t risk her learning his secret. Debra thanks Peter for his quick thinking. Peter notices that Dr. Connors isn’t there, so he heads to his house, hoping to see him safe in his home.

When he arrives, he startles Margaret Connors, who is frightened by Spider-Man. Soon enough, the Lizard arrives, and Spider-Man and he have a quarrel. After the fight, the Lizard runs off. Spider-Man hears Billy, Dr. Connors’ son call the Lizard “daddy”, which he questions them about. Margaret explains that Dr. Connors used the neogenic recombinator, along with a special reptilian serum that was supposed to rejuvenate his lost arm. The experiment was a success, though he began to become lizard-like, until he became a full-blown, humanoid lizard. Spider-Man hears a disturbance nearby, and sees what it is. It turns out to be Eddie Brock, who was listening in on the conversation, and plans to alert Jameson of his findings. Spider-Man webs him to a light post, as he refuses Eddie to ruin the Connors’ life. The Lizard returns and the two begin their battle once more. Spider-Man is knocked unconscious for a short period; though that gives the Lizard just enough time to take Margaret into the sewer. Spider-Man takes Billy to his aunt, and then heads into the sewer after the Lizard. While tracking the Lizard in the sewer, Spider-Man makes various references about many other Marvel superhero teams, such as the Fantastic Four, the Defenders, and the Avengers, and how they wouldn’t be caught dead in the sewer. He climbs through a drainage pipe, which sends him down at a fast speed due to it being inverted. Once he crashes through the gate at the end of the pipe, he shoots a line of webbing to catch himself before he falls to his death at the bottom of the area. He begins wall-crawling across the wall when he’s grabbed through a drain gate. The arm belongs to the sewer worker who was abducted in the beginning of the episode. The worker explains the Lizard’s plot to turn the entire human race into humanoid lizards like him. Spider-Man heads off, though the weak worker asks Spider-Man if he’ll come back. Spider-Man responds saying he’s a superhero.

We now see Margaret Connors, who is sitting before the Lizard, as he plans to use the neogenic recombinator on her to make her the first of billions of humanoid lizards. Margaret tries to convince him to stop his plans, which is a success after she reminds him that he has a son, and that he wouldn’t want to turn him into a lizard as well.

Spider-Man drops in, causing him and the Lizard to engage in combat. In an attempt to thrash Spider-Man with his tail, the Lizard mistakenly tail whips his wife. Spider-Man takes his mourning as an advantage, and tries to use the neogenic recombinator on him to reverse the process. He is knocked into the water, along with the neogenic recombinator, which the two fight to get a hold of. Spider-Man finally obtains it, then turns it on, thus returning Dr. Connors to his original state.

The scene changes to J. Jonah Jameson getting out of a car, along with Eddie Brock and a few reporters from the Daily Bugle. Jameson is curious as to what this is about, as Peter already brought him photographs of the Lizard. Eddie Brock claims that Dr. Connors is the Lizard, and that should he be incorrect, he’ll eat today’s issue of the Bugle. Dr. Connors answers the door with Billy and Margaret, in which Jameson asks Eddie if he’d like the newspaper “cooked or raw”.
