In Limestone, a teenage girl, Kristen, goes to the Black Rose bar and finally find the boy she's there to meet: Robert. They sit and discuss her poetry, and Robert admits he's impressed by them. He insists that he knows her, and asks to see some of her work. As she gets it out, she gives herself a paper cut and Robert turns away, insisting he has to go. He runs off before Kristen can stop him. However, later, he returns and admits he couldn't stay away. Robert then says that he has to show her something. He pulls up his lip and reveals that he has fangs. Kristen, impresses, says that she knows it and starts to bare her throat. She wonders if it will hurt as she bares her neck, but he offers to show her his world instead. He takes her to an abandoned building and a muscular vampire leaps out. He compliments Robert, and the two vampires attack her.

As Dean waits for Sam at the Limestone police station, he calls and tells Lisa that he's only an hour away, and he'll stop by once they've solved the case. She tells him to get him as soon as possible, and to stay safe. Sam comes out with information on the seven girls that have recently gone missing. All are in the late teens and talented, They decide to talk to the parent of the most recent girl who has disappeared, He lets them search Kristen's room, and they discover that she was a fan of bestselling vampire books. Sam finds her laptop and eventually manages to figure out her vampire-related password, while Dean grumbles about vampires being romanticized. They find emails from Robert to Kristen where he describes himself as a vampire. They figure that vampire-obsessed girls would be easy prey for vampires, and discover that Kristen went to the black Rose.

That night, a delivery driver is ambushed and vampires kill him and take his cargo. They drive away, leaving the driver dead... next to his blood donor van.

Once Samuel gets word of the raid, he informs Sam that vampires are involved. They wonder why the vampires need a blood van if they have victims, and Samuel suggests they find the nest. The brothers go to the Black Rose, but find it difficult to pick out just one vampire-looking individual. Their two likely targets head outside and the Winchesters split up to follow them. Sam follows his target into a storage room. When the vampire comes at him, Sam decapitates it with a single knife blow.

Dean's target gets his girl victim up against the wall. Dean stops him, but realizes that he's a fake vampire, and the boy admits that he pretends to be a vampire to get laid. He sends the boy off, but the hulking vampire, Boris, grabs him and throws him across the alleyway. Dean tries to fight back without success, and Sam arrives. He stops and simply watches as Boris opens a vein and feeds the blood to Dean. Sam finally draws a knife and attacks Boris, who runs away, laughing.

Back at the motel, Dean is overwhelmed by the sights of sounds impacting on his newly heightened senses. Dean wants Samuel to kill him when he arrives, and figures that Sam won't do it. However, he's able to hear Sam's heartbeat and wonders why he isn't more upset. Dean insists that they have to kill him before he feeds on someone, then goes into the bathroom and examines his new fangs in the mirror. A few minutes later, Sam realizes that Dean is taking too long and opens the door, and discovers is brother has gone out through the window.

Lisa wakes up and is surprised to find Dean in her room. He tells her that he appreciates everything she and Ben have done for her, and then starts to go. Lisa demands an explanation and Dean says that he can't bring crap home with him. He starts to approach her, growing fangs, but then pulls away and runs out into the hallway. Ben comes out and Dean warns him back, and then shoves him into the wall and runs outside when he ignores his warning.

Samuel arrives at the motel and syas he has to do the right thing, and Dean steps out of the shadows. He explains that he went to visit Lisa, but assures Samuel that he didn't feed... yet. Dean tells Samuel to kill him, but Samuel informs him that there's a cure that he got from his own grandfather's journal. However, he warns Dean that it won't work if he drinks any human blood. One of the ingredients is the blood of the vampire who turned Dean. Dean knows where the nest is using his heightened sense, and figures he can go in posing as a vampire, use dead man's blood to take out Boris, and get the blood. Samuel gives him a syringe of dead man's blood to use and wishes him luck. Once Dean leaves, Samuel asks Sam why he didn't remember them discussing the cure a few months ago. Sam claims to have no memory of the conversation, but Samuel wonders if he set up Dean to infiltrate the nest and track down the alpha vampire they're looking for. Sam questions Samuel's motives and insists that he's just glad they can cure Dean.

Dean goes to the abandoned building and sneaks in. He runs into Robert, who welcomes him into the nest and says that Boris told them to expect Dean. Robert offers him blood, and Dean claims that he killed enough people that he's not hungry. His host says that they're not allowed to kill humans anymore, because of "company policy." Robert then takes Dean to a central chamber. Some vampires are caged, feeding on blood, and Boris is coaching Kristen in how to attract vampire fans. He sends her to bring in another one, and then greets Dean. He admits he wondered if the hunter, Sam, got Dean. Dean professes ignorance of hunters, and Boris boasts that they can easily handle them. Boris then asks Dean how old he is, and admits that he's over 600 years old. He explains that it's the best days to be a vampire since the public is obsessed with them, and Dean will be one of his "recruiters" and bring them in. The girls will go out and bring in more guys, they'll be converted, and so on. When Dean asks if it's Boris' idea, Boris says that it's their father's idea and looks skyward.

Boris offers Dean the private tour. Once he turns his back, Dean gets out the syringe and prepares to stab him, but a single drop of blood spills and Boris hears it. He disarms Dean and prepares to kill him, but there's a sudden muttering of voices. First Boris and then the captive vampires collapse, muttering "father." Finally, Dean collapses as well. Unconscious, he dreams of an ancient vampire spreading his blood throughout the country, and twin vampire girls in an old house.

Dean wakes up in the central room as the vampires move in on him. He draws his knife and starts killing vampires, decapitating Robert.

Outside, Samuel and Sam arrive. A vampire comes flying out of the window and lands on their truck. Dean continues to hack his way through the vampires. and finds himself face-to-face with Boris. The head vampire warns him that he hasn't stopped anything, and that the situation is bigger than both of them. He then gestures for Dean to attack him. A few minutes later, Sam and Samuel arrive and find Dean standing over the decapitated Boris. He tells them that he's good.

Back at the motel, Samuel finishes the cure while Sam asks Dean what he saw. Dean tells him to back off and drinks the cure, and then starts vomiting. He briefly transforms into a full-fledged vampire, remembering everything that happened, including Sam watching as Boris fed him his blood. When he snaps out of it, Dean is once more human.

The next morning, the Winchesters pack and Sam asks Dean what he saw. Dean explains that the alpha vampire is sending them psychic messages and building an army of vampires. He also tells Sam and Samuel that the hunters don't scare them anymore,

Dean tries to call Lisa, who refuses to pick up. He can't bring himself to leave a message. Sam expresses his sympathies, and Dean sarcastically notes that he can count on Sam no matter what. Sam agrees, but Dean isn't convinced.
