A man is working at home when he hears someone moving around outside. He calls to what he assumes is his wife but gets no answer. Assuming he imagined it, the man goes back to work, but someone or something picks him up, throws him across the room, and then cuts off his hands and feet and then carves a symbol into his chest.
Sam is driving to the town where the murders take place, and Dean wakes up. He takes a drink from Bobby’s old flask and says that he’s dealing with their friend’s death by honoring his memory. Dean figures that it’s better than how Sam is coping, by going off on a hunt. The brothers have confirmed that four men have died in the same town, and in each case their attacker used superhuman strength and cut off their hands and feet. Posing as FBI agents, the Winchesters talk to the coroner, Eddie, who informs them that one of the victims managed to bite his killer. However, the DNA tests indicated that the flesh sample wasn’t human. As the brothers leave, Dean suggests that his brother go on ahead while he checks out the area. Sam realizes that Dean wants to go to a bar and drink, and Dean doesn’t even both denying before walking off.
Dean finds a local watering hole, the Cobalt Bar, and immediately joins up with a woman, Lydia. He claims to be an investment banker and it’s clear that Lydia is into him. She soon invites Dean back to his apartment and they end up having sex.
Jerry Price is at home in his apartment when there’s a knock at the door. He answers it and says hello to the person. The visitor throws him across the room and then kills him just like the other four men.
The next morning, Dean meets up with Sam and they go to check the new murder. Sam suggests that they find an expert since he couldn’t identify the symbol on the corpse. Inside they talk briefly to the head detective, Charlene Penn. Eddie brings them up to speed and Sam notices a man outside. He talks to the person, who identifies him self as Jerry’s friend and next-door neighbor. He knows that Jerry had an affair and cheated on his wife Ann. Outside, Sam briefs Dean and they figure that the wife wasn’t involved. When Dean reaches for his flask, he discovers that he left it at Lydia’s place. He calls her but she abruptly cuts him off. Sam, listening in, is amused that Dean is being brushed off by a one-night stand this time. Meanwhile, a very pregnant Lydia puts the phone away.
A short time later, Lydia is at a building surrounded by midwives. They help her deliver the body, insisting that the pain she’s feeling is an honor. The woman in charge, Madeleine says that they’ll call the girl child Emma and then calls for the next woman.
Dean and Sam go to see a local archaeology professor, Morrison. Morrison is intrigued by the symbol but would like payment. The best they can do is put in a good word with the IRS. Morrison is less than impressed and asks for a green card for his housekeeper in return for his help. As they leave, Dean complains that they have to turn to Morrison now that Bobby is dead. When he realizes that Lydia hasn’t called him back, Dean tells Sam that he’s going to her place to recover the flask.
Lydia greets Dean but isn’t sure what his name is. As she goes to get the flask, Dean notices that she has a baby in the next room. He tries to talk about the 2-year-old, but Lydia isn’t interested. Dean takes a call from Sam and Lydia checks on her daughter. As Sam says they’re needed at the crime lab, Dean overhears Emma clearly asking her mother who their visitor is.
At the morgue, Sam asks to check out the new victim. Charlene is there and wants to know why the FBI is involved. Eddie suggests that it’s a cross-state crime and notes that there have been other victims in different states. Once Charlene leaves, Sam asks for the files on the other cases that Eddie has uncovered. He also finds a matchbook for the Cobalt Bar among the dead man’s possessions. The other local victims were there as well, and victims in other states were men who had affairs and went to bars.
Dean watches Emma’s house and sees Madeleine and another woman go inside. Sam calls to tell Dean about the other murders, each series two years apart from the others. He also tells Dean that some of the local victims were seen at the same bar Dean was at. The women emerge with Emma, and Dean is surprised to see that she is now five years old. Lydia tells Emma to make them all proud of her and then Madeleine leaves with the girl. Dean follows them to a warehouse and then goes back to the hotel to tell Dean what he saw. Sam figures that Dean is obsessed and doesn’t believe him, and their argument is interrupted when Morrison calls them.
At the warehouse, Madeleine gathers Emma and the other children. She and the other adult women perform a ritual which involves praising the one who created them by hunting and killing, and offers each child a piece of meat. Madeleine tells Emma to eat, and the girl hesitates briefly before consuming it.
Morrison gives a presentation on the symbol, explaining that it is Greek and belongs to Harmonia, the goddess who mated with Ares to produce the Amazon. The Amazons only used men for procreation and then killed them, cutting off their hands and feet.
Charlene, an Amazon, calls Madeleine and tells her that she’s identified them as hunters. Madeleine says that they’ll dispose of both brothers.
Back at the hotel, Dean tries to find Bobby’s book on Grecian mythology. Sam checks the Internet and confirms that the Amazons bargained with Harmonia. She turned them into monsters but the lore isn’t clear if there is a special way to kill them. The Amazons’ children grew to adulthood incredibly fast, creating a rapid army. The mating cycle is every two years, accounting for the pattern of killings. They realize that Dean is a father and Emma is his daughter, despite the fact that he used protection.
Madeleine addresses the now teenage children. She brands each one to indoctrinate them into pain, both enduring and giving it. Emma winces in pain and Madeleine tells her that courage is everything.
At the hotel, the Winchesters go through Bobby’s files. Sam wonders why Lydia hooked up with Dean, and Dean admits that he posed as a rich investment banker. One piece of paper from the files move, even though there’s no wind in the room. Sam scans for ghosts but the local power lines give false readings. Dean wonders if Bobby’s ghost is with them, but Sam angrily insists that they burned him and he couldn’t be back. Sam picks up the paper and Dean suggests that it could be useful. It’s in Greek and Sam realizes that they have to show it to Morrison. He tells Dean to lock the door and stay where he is while he talks to the professor.
At the university, Morrison reluctantly looks at the paper and Sam agrees to remove the wiretap from his phone.
There’s a knock at the hotel door and Dean answers it. It’s the teenage Emma, who asks for his help because Dean is the only person that she can trust because he’s her father. She explains that the Amazons have been watching him ever since Lydia got pregnant, and she escaped when the shifts changed. Emma insists that she doesn’t want to be like the Amazons and Dean invites her in. She asks him to get her away from them, and claims that Lydia said he was a good man. Dean says that he isn’t and doesn’t know what to do, but Emma says that she hasn’t had a normal life and it’s her last chance to have one.
Morrison dwells on the quality of the paper. Sam gets him to focus and Morrison confirms that according to the ritual, the child must kill her own father.
Emma says that she’s starving and asks if Dean will help her.
As Sam leaves the archaeology building, Charlene intercepts him. She grabs him and he sees her Amazon brand. She throws him down the hallway and draws a knife, but Sam shoots her in the chest, killing her. He checks his phone but discovers that it was smashed in the fight.
As Sam drives across town, Dean goes to the refrigerator to get some food for Emma. As soon as his back is turned, she produces a knife and prepares to kill him. Dean turns, his gun drawn, and figures that he knew she was up to something. Emma insists that she has to kill him because of what she is, and points out that if he could bring himself to kill her, he would have already done it. She appeals to his sentiment as Sam arrives and overhears them talking. When Dean doesn’t pull the trigger, Sam bursts in and prepares to shoot her. She begs Dean to save her and Dean hesitates, and Sam shoots her in the chest.
The brothers go to the warehouse but discover that the Amazons have already escaped and taken everything with them.
Later, an angry Sam is driving away with Dean in the passenger side. Dean tries to assure him they’ll be ready for the Amazons, but Sam is mad that Dean couldn’t bring himself to shoot a monster. He points out that Dean killed Amy, and his brother says that he was going to kill her. Sam warns his brother that his heart isn’t in it and he’s going to get them both killed. Dean insists that Sam is just as screwed up as he is, but Sam just asks him not to get killed. The best Dean will do is say he’ll do what he can.
