Professor Cox approaches Crawford Hall and finds an attractive girl waiting out front. She says that she's in one of his classes and asks to meet with him. Cox agrees and they go to his office, and she admires his book on modern morality. The girl finally admits that she's not one of his students and starts to go, but Cox tells her that what she's feeling is natural. Cox talks about respecting her too much to take advantage of her and then kisses her. The girl kisses him back and Cox realizes that her face is rotting away. She asks if he likes her anymore and advances on him.
Later, the university janitor is locking up. As he walks outside, he hears a thudding noise behind him and discovers Cox lying on the steps in front of the building, dead.
One week later, Dean and Sam are holed up in a motel and Sam complains that he doesn't have his computer. Dean tries to irritate him, and then complains that the Impala is screwed up. Sam starts to explain he had nothing to do with it, but a knock on the door interrupts him. It's Bobby, who is there in response to their call to check out the case they're working. They explain that they investigated Cox's death, posing as reporters
Sam talks to Jen and Curtis, and Jen talks about an urban legend about how 30 years ago, a girl was having an affair with a professor and then killed herself when he broke it off. The legend says that now the girl haunts the campus, and she jumped from room 669. Curtis doesn't believe it and laughs at her. Sam goes to the bar where a boozy Dean is drinking purple nurples and is trying to pick up a drunken girl, Starla, by claiming he's an astronaut pilot.
Dean interrupts to say it didn't happen like that, but the girl was classy and he discussed ghost stories with her.
Dean talks to the classy girl over purple nurples and romantic dialogue. She finally can't resist kissing him. Sam comes over and rudely interrupts them, insisting it's a serious investigation.
Sam insists he doesn't sound like that. Bobby realizes that something is up, but Sam insists they've just been together too long in tight quarters. He then explains that they went to the scene of the crime.
The brothers talk to the janitor, who lets them into the office. He notes that the professor jumped out his fourth-story window, and he's the one who found Cox dead. The janitor claims that Cox wasn't alone. Meanwhile, Dean stuff his face with peanut brittle.
Dean objects that he didn't have that many mints.
The janitor explains that Cox was with a young lady, and she wasn't the only one. Sam confirms that there's no 669 in the room. Back at the motel, Dean suggests they check out the history of the building. Sam boots up his computer and discovers that Dean froze it on a porn site.
Bobby confirms that they didn't find up anything on a ghost, and Dean admits they're not sure. He says that they didn't see the next part, but it's weird.
Curtis is walking past Crawford Hall when he hears something above him. A bright light shines down from above and lifts him into the sky.
Bobby doesn't believe it, and Sam and Dean admit that it sounds pretty unlikely. They decided to talk to Curtis to make sure.
Curtis is downing drinks at the bar and tells the brothers that they won't believe him. However, he goes ahead and tells them that once the aliens captured him, they probed him repeatedly... and then slow-danced with him.
Bobby doesn't believe it, but the brothers assure him that Curtis may have been telling the truth.
The Winchesters check the grounds outside the hall and find a patch of blasted grass. They then talk to one of Curtis' frat brothers, and Dean describes what happened to Curtis. The frat brother says that he didn't see anything, and Sam hugs him and consoles him, according to Dean.
Sam objects, saying he never did anything like that.
The frat brother tells them that Curtis was their pledge master and got off on it, and he figures now Curtis knows how they feel. Back at the motel, Dean figures that both victims got what was coming to them. Sam realizes that his laptop is gone, and Dean says that he has no idea where it is. His brother doesn't believe him and starts yelling at him for messing with his stuff.
Bobby confirms that Dean didn't take the laptop, and Sam insists that he didn't take the computer. The brothers go on and say that there was one more victim, a research scientist involved with animal testing.
The research scientist is leaving Crawford Hall and sees a glint of gold from a nearby sewer grate. He reaches in to grab it, and something bites his arm. Later, the brothers break into the morgue and find his dismembered body. Sam examines the remains and finds an alligator scale. Dean insists the legends are just legends, but Sam isn't so sure and decides to call in Bobby. They search the sewers and Dean discovers that someone has punctured all four tires on the Impala. There's a money clip on the ground next to the Impala, with Sam's initials on it. He goes into the motel room and accuses Sam of playing the prank, but Sam insists that he didn't do it. He wants his money back and tries to grab it, and they end up wrestling on the bed.
Dean explains that Bobby arrived an hour later. Bobby tells them that the other one didn't do the pranks, He explains that they have a trickster on their hands, and they create chaos and mischief. It's been playing them to keep them off its tracks. Bobby explains that tricksters are immortal demi-gods that can create and vanish anything they want. Tricksters target the high and mighty to knock them down a peg, and can look like anything they want, typically human. Dean and Sam realize what human has been in the vicinity every time.
The janitor is in his apartment reading tabloids and feeding his dog from a table filled with candy. When he realizes something is missing, he summons two beautiful girls in lingerie.
The next day, the Winchesters return and claim they have to check their offices. Sam goes back to get something, and then breaks into the locker room and searches the janitor's locker. He finds tabloids but nothing else. Outside, the brothers meet back up and Sam admits he found nothing else. Dean notes that Bobby told them that tricksters have a sweet tooth, but Sam didn't find any candy. They start arguing again, unaware that the trickster is watching them from the window. Sam tells Dean to wait until he finds some evidence and storms off.
That night, Dean is still waiting outside Crawford Hall and there's no sign of Sam. He finally goes inside, removing a wooden stake from his jacket. When he hears music, Dean goes into the auditorium and finds the Trickster's two women on a bed, waiting for him. They offer to give him a massage, and Dean admits that he's tempted. The Trickster appears in the audience and explains that the women are a peace offering. He insists that his victims got what they deserved, but he likes Sam and Dean. All he wants to do is move on to the next town. Dean says that they can't, and the Trickster points out that he shouldn't have come alone. Sam comes in, and the Trickster realizes they staged the fight. He summons a masked chainsaw-wielding maniac to attack Sam, while the women attack Dean. Bobby comes in to help Sam, while the women throw Dean into the seats. Sam tosses him a stake, and Dean impales the Trickster. The women and the maniac disappear as the Trickster collapses, dead.
As the trio get into the Impala, Sam tries to apologize, and Dean says it's fine. They drive off... unaware that in the auditorium, the real Trickster has emerged from the shadows to make his fake body disappear.
