Sam wakes up in a hotel room while Heat of the Moment plays on the radio alarm. Dean goes through his (long) morning ritual and they go to the local diner. Dean would rather be looking for Bela, who has the Colt, but with no leads Sam's decided they should check out the reported disappearance of a Professor Hasselback who visited the local Mystery Spot, a gravity-defying tourist attraction. The waitress, Doris, spills a bottle of hot sauce on the floor. Outside, the guys walk by a dog and an attractive blond bumps into Dean. They walk by two men moving a heavy desk and decide to break into the Mystery Spot that night. They don't find anything unusual, but the owner, Mr. Carpiak, comes after them with a shotgun. When Dean tries to calm him down. Carpiak shoots him down dead.

Sam wonders if he had a vivid dream but soon realizes that it's the exact same day, but Dean doesn't remember anything. This time Sam catches the bottle of hot sauce. They go outside and run into the dog, the blond, and the movers. Sam suspects that something happened at the Mystery Spot but suggests they go right away so Dean doesn't get shot. Dean agrees... and gets hit by a car crossing the street.

Sam convinces Dean they're in a time loop and this time they make it to the Mystery Spot. They meet with Carpiak, who after giving them a long spiel about the Mystery Spot being a family heirloom, finally admits he bought it at auction a month ago. With no further answers, they head back to the hotel... and Dean gets crushed by a falling desk.

This time Sam keeps Dean in the hotel room... but Dean falls and kills himself in the shower. They keep going through the same loop, but Dean manages to get food poisoning from a bad taco, electrocutes himself with a shaver, and ends up accidentally hacked to pieces when Sam tries to tear apart the walls of the Mystery Spot to get answers.

After a hundred loops, Sam once more tries to explain to Dean what's going on. This time when they go outside Dean goes after the blond woman and discovers she's Hasselback's daughter, handing out "Missing" posters. When Sam runs after her, Dean tries to pet the dog... which kills him.

The next time in the loop, Sam explains that he found out that Hasselback was a pompous jerk who debunked supernatural tourist attractions. As they prepare to leave, Sam notices a local judge has used a different syrup on his pancakes. Suddenly the loop starts again, even though Dean didn't die. Sam takes Dean to the diner again and then goes after the judge when he leaves. He holds a stake to the judge's throat and demands answers, insisting there's only one creature powerful enough to create a time loop. The "judge" finally confesses that he's not just a Trickster, but the Trickster that they believed they'd killed. The Trickster disposed of Hassleback as a jerk and then noticed the Winchesters when they arrived in town. He admits he thinks the whole thing is funny but he's also teaching Sam a lesson. Sam threatens to kill him unless he breaks the loop, and the Trickster agrees. However, Sam doesn't believe him and the Trickster snaps his fingers and...

Sam wakes up in a hotel room while Back in Time plays on the radio alarm. it's now the next day, Wednesday. Sam tries to get them out of town, but a local homeless man comes up to Dean in the parking lot and shoots him dead. This time, Sam doesn't wake up.

For the next three months, Sam travels across the country looking for the Trickster and killing demons and vampires along the way. Bobby tries to get hold of him repeatedly but Sam doesn't return his calls. Finally Bobby calls and says he's found the Trickster at the spot where he last used his magic. Sam arrives but Bobby warns they'll need a gallon of fresh human blood to summon the Trickster. Sam shrugs and offers to kill someone to get it, and a disappointed Bobby reveals it's a ruse to lure Sam in so they can talk. Sam refuses and Bobby offers himself as a sacrifice, insisting he's too old and this way they can get Dean back. Sam prepares to cut his throat... then stabs him through the chest with a stake, the only way to kill the Trickster. It's not the Tricker, but just another of its allusions. The "corpse" disappears and the real Trickster appears. The Trickster warns that he's trying to teach Sam a lesson, and that Dean is his one vulnerability. Sam begs the Trickster to bring Dean back and the Trickster, now bored, agrees to do so. However, he warns that something bad will happen. He snaps his fingers, and...

Sam wakes up on Wednesday, and Dean is still alive. As they head out, Sam contemplates the room... and leaves with his brother, reunited.
